Effects of Pseudomonas aeruginosa Lectin, PA-1 on Cytokine Gene Expressions from Human Peripheral Bleed Mononuclear Cells




Abstract
The gene expressions of 5 cytokines(IL-1, IL-2, IL-6 TNF¥áand IFN¥ã) from human peripheral blood mononuclear cells(PBMC) stimulated with Pseudomonas aeruginosa lectin(PA-1) were investigated by using reverse transcription polymerase chain
reaction(RT-PCR).
PBMC(1¡¿10E6 cells/ml) isolated from healthy volunteers were stimulated with PA-1 for various times(1, 8, 24, 48, 72, 96 and 120 hours) and various concentrations(10, 50, 100ug/ml) for 24 hours. After each of the various stimulated times and
concentrations, total RNA was isolated by RNAzol B method and assessed for various cytokines mRNA by RT-PCR.
Effect of PA-1 doses on the induction of cytokines showed differently among the cytokines. The induction of IFN¥ãmRNA were PA-1 dose dependent, but the induction of IL-6 mRNA was expressed evenly regardless of stimulated doses.
Cytokine gene expression in various times stimulation of PBMC showed diverse patterns among the cytokines. Significant increased expressions of all cytokines mRNAs were observed at 24hours time point. And about 24 hours time point, the expression
patterns of five cytokines showed diversely. The inductions of IL-6 and TNF¥ámRNA were more rapid and the induction of IL-2 mRNA was late compared with other cytokines. The expressions of IL-1 and IL-6 mRNA were maintained to 96 hours time point.
whereas the expressions of other cytokines mRNA were maintained to 48 hours.
These results suggest that PA-1, as an inducers of cytokines, can elicits a detectable cytokine on mRNA from the PBMC within the first few hours of stimulation and maintains the production of cytokines during a few days.
